Treatments for childhood cancers are increasingly successful with cure rates approaching 80%, but success often comes with a downside for the surviving men: the cancer treatments they received as boys can leave them sterile as adults. Now, a research team led by Ralph Brinster of the University of Pennsylvania School of Veterinary Medicine has completed a 14-year experiment that gives hope for a technique that could restore their fertility.

Brinster is the Richard King Mellon Professor of Reproductive Physiology at Penn Vet and was recently awarded the National Medal of Science for his lifetime of research on the genetics of the mammalian germline, the cells that give rise to sperm and eggs.

In his most recent research, Brinster collaborated with fellow members of the Department of Animal Biology at Penn Vet, with members of the Department of Cell and Developmental Biology at Penn's Perelman School of Medicine and with the Penn Bioinformatics Core.

Their study was published in the journal Human Reproduction.

For males, fertility begins with spermatogonial stem cells, which are present at birth, embedded in the basement membrane of the testes' seminiferous tubules. As a boy approaches puberty, these cells begin to make daughter cells that eventually become sperm. While they normally continue this process throughout a post-pubescent man's life, factors like radiation and chemotherapy drugs can destroy them, rendering him sterile.

About 1 in 3 boys surviving childhood cancer will be in danger of having severely decreased fertility as an adult; as many as 1 in 5,000 men of reproductive age currently suffer this serious quality-of-life problem as a result. Adult men who undergo cancer treatment that might damage their fertility can preemptively freeze their sperm, an option not available to pre-pubescent boys. "There are a number of places, including at the Children's Hospital of Philadelphia," Brinster said, "that are already freezing cells for patients to use later, with the expectation that the necessary culture system and implantation techniques will be developed. A logical question for patients to ask is, How do we know that, after 10 years or more of being stored, these cells are any good? That's what our study addresses."

The techniques for extracting these cells and re-implanting them have been developed, so a critical question for researchers was whether spermatogonial stem cells could survive the decade-plus period they might need to remain frozen.

Fortunately, Brinster had a large collection of cryopreserved spermatogonial stem cells stored in the mid-'90s. The collection consisted mostly of cells taken from mice but included a small number of rat, rabbit and baboon samples. Between their age and variety, these samples represented an important resource to address questions regarding long-term cryopreservation of spermatogonial stem cells, before the technique could be used in humans.

After being thawed, the spermatogonial stem cells of rabbits and baboons were labeled with a fluorescent dye so that the researchers could track where in the testes they would eventually migrate and embed once they were implanted in mice. This was a critical step for the rabbit and baboon cells, which lacked appropriate experimental recipients; once implanted in mice, the studies showed that the cells migrated correctly and embedded in the recipient testes' basement membrane, indicating their viability.

A complete functional test could only be done for the mouse spermatogonial stem cells, as they could be implanted into mice and tested to see if they could produce sperm, and whether that sperm could lead to healthy offspring. After demonstrating that the cryopreserved cells implanted in the right region of the testes and underwent normal spermatogenesis, the researchers extracted the sperm and successfully fertilized eggs in vitro.

One of the recipient mice was also placed with females and successfully sired offspring. The offspring resulting from eggs fertilized in vitro as well as those resulting from natural mating all produced normal appearing young. Most importantly, the offspring arising from the cryopreserved spermatogonial stem cells appeared free of genetic damage.

This experiment clearly points in the right direction for the feasibility of a similar fertility treatment in humans.

"Human and animal spermatagonial stem cells have been successfully frozen for short periods of time, but this is completely different," Brinster said. "Here we had cells frozen for over a decade that implanted in the right place and made sperm, and that sperm made offspring without apparent genetic defects."

(AP) ? Guatemala's president proposed Saturday that the United States and other "consumer" countries pick up the tab for the cost of drug seizures.

Otto Perez Molina made the comment during a Central America meeting that he called to discuss his earlier proposal to legalize drugs, a meeting that drew only two of his counterparts from the region.

"For every kilo of cocaine that is seized, we want to be compensated 50 percent by the consumer countries," he said.

The Guatemalan leader said the United States has a "responsibility" because it has one of the highest rates of drug use.

Drug cartel activity has been increasing in Central America, both as a smuggling route and a base for operations.

Perez Molina said the fight against drug trafficking has failed and governments need to look for alternatives.

The presidents of Panama and Costa Rica did attend Saturday's meeting in Guatemala, and two of the other four Central American nations sent lesser representatives.

The office of Salvadoran President Mauricio Funes suggested setting a date for another meeting, saying other nations have expressed interest in participating.

Perez Molina first made the drug legalization proposal in February. It has received lukewarm response in the region and opposition from the United States.

He said Saturday that legalization of drug use should be accompanied by a system to regulate the production and consumption of drugs. He also suggested the region decriminalize drug trafficking and instead set up a specific transit corridor with border controls for the registration of drug shipments.

Army sergeant charged in Afghan massacre

WASHINGTON (AP) ? Charges filed Friday against Army Staff Sgt. Robert Bales reflect the horror of the crime: 17 counts of premeditated murder, more than half of them children, during a shooting rampage in southern Afghanistan. But while Afghans are calling for swift and severe punishment, it will likely be months, even years, before the public ever sees Bales in a courtroom.

One only has to look at two recent and similarly high-profile cases to see that the wheels of military justice turn slowly.

It's been nearly 29 months since an Army psychiatrist, Maj. Nidal Hasan, allegedly killed 13 and injured two dozen more at Fort Hood, Texas. His trial is scheduled to begin in June. And it's been 21 months since the military charged intelligence analyst Bradley Manning with leaking hundreds of thousands of pages of classified information. It took nine months before he was deemed competent to stand trial.

The Bales case is likely to be equally complex, involving questions of his mental state and the role that the stresses of war and possible previous head injuries may have played in his alleged actions. Most of the eyewitnesses are the Afghan villagers and survivors who may be brought in for the trial.

The military on Friday charged Bales with 17 counts of murder, six counts of attempted murder and six counts of assault in the March 11 pre-dawn massacre in two southern Afghanistan villages near his base. The father of two from Lake Tapps, Wash., was officially informed of the 29 charges just before noon at the U.S. military prison at Fort Leavenworth, Kan., where he is confined.

The maximum punishment for a premeditated murder conviction is death, dishonorable discharge from the armed forces, reduction to the lowest enlisted grade and total forfeiture of pay and allowances, according to Col. Gary Kolb, a spokesman for U.S. forces in Afghanistan. The mandatory minimum sentence is life imprisonment with the chance of parole.

The charges offered few details of what happened that night. But the 38-year-old soldier is accused of walking off his base with his 9mm pistol and M-4 rifle, which was outfitted with a grenade launcher, killing four men, four women, two boys and seven girls and burning some of the bodies. The ages of the children were not disclosed.

In the most detailed descriptions of the shootings to date, the charges say Bales shot a young girl in the head, a young boy in the thigh, a man in the neck and a woman in the chest and groin. The documents also say that he "shot at" another girl and boy, but apparently did not hit them.

The attack occurred in the Panjwai district of Kandahar Province, the spiritual birthplace of the Taliban. The dead bodies were found in Balandi and Alkozai villages ? one north and one south of the base.

Members of the Afghan delegation investigating the killings said one Afghan guard working from midnight to 2 a.m. saw a U.S. soldier return to the base around 1:30 a.m. Another Afghan soldier who replaced the first and worked until 4 a.m. said he saw a U.S. soldier leaving the base at 2:30 a.m. It's unknown whether the Afghan guards saw the same U.S. soldier. If the gunman acted alone, information from the Afghan guards would suggest that he returned to base in between the shooting rampages.

It also is not known whether the suspect used grenades, Kolb said. The grenade launcher attachment is added to the standard issue M-4 rifle for some soldiers but not all, he said. Bales was assigned to provide force protection at the base.

The case against him is the worst allegation of killing of civilians by an American in Afghanistan and has severely strained U.S.-Afghan ties at a critical time in the decade-old war.

Not addressed in the charges are suggestions that Bales may have been drinking. On Friday, a senior U.S. defense official said Bales was drinking in the hours before the attack on Afghan villagers, violating a U.S. military order banning alcohol in war zones. The official spoke on condition of anonymity to discuss the charges before they were filed.

U.S. officials, however, have said that additional charges could be filed as time goes on. Bales' civilian attorney, John Henry Browne, said Friday the government will have a hard time proving its case and that his client's mental state will be an important issue. Bales was on his fourth tour of duty, having served three tours in Iraq, where he suffered head and foot injuries.

Meanwhile, an attorney for Bales' wife said the couple have spoken by telephone twice since he was detained. Attorney Lance Rosen said Bales called his wife, Karilyn, first from overseas shortly after the March 11 pre-dawn massacre, then from Fort Leavenworth on Wednesday.

Rosen said Bales and his wife were warned the call Wednesday would be monitored and told they would only have 10 minutes to talk. They talked about family matters and "reaffirmed their love for each other," Rosen said. He also said the family has set up a defense fund to help pay for Bales' legal fees.

The charges set in motion what will likely be a lengthy military justice process.

"Usually in military and civilian criminal cases, delay accrues to the benefit of the accused. So there will be some maneuvering to put it off a while," said Gary Solis, retired Marine prosecutor and adjunct law professor at Georgetown University. "I think it's many months before we're going to see this thing go to trial, if it ever goes to trial."

Bales may face what the military calls a "sanity board," to determine his mental state at the time, and whether he is competent to stand trial.

Since Bales was assigned to a unit based at Joint Base Lewis-McChord in Washington state? the 2nd Battalion, 3rd Infantry Regiment of the 3rd Stryker Brigade Combat Team of the 2nd Infantry Division ? the charges were sent Friday to a special court-martial convening authority, the 17th Fires Brigade, an artillery unit at the post. Lewis-McChord spokesman Lt. Col. Gary Dangerfield said officials at the post will have the legal responsibility of trying and managing the case against Bales, but it was not clear where the proceedings would actually take place.

Maj. Christopher Ophardt at Lewis-McChord said the preliminary hearing may not take place for a few months and it would likely be several more months after that before a trial would begin ? perhaps two years from now depending on motions and other pretrial actions.

Jeffrey Addicott, who previously served as the senior legal adviser to the U.S. Army's Special Forces, said the military justice system typically moves faster than civilian courts. But he said the international political implications may cause this case to drag on for years.

"When we have these high-publicity cases, the military becomes like a deer in the headlights. They have just one speed: extremely slow," Addicott said, adding that he expects the defense team to use all tactics to slow the process.

According to military lawyers and experts, now that the charges have been filed, the next step is for the military is to decide whether there is enough evidence to refer the charges to a preliminary hearing. That hearing, what the military calls an Article 32, would determine whether there's probable cause to believe a crime was committed and that the person charged did it.

Once the Article 32 is over, the hearing officer recommends to a general officer whether or not the case should go to trial. That general officer then decides whether the case should go to a court-martial in front of a military judge.

Throughout the proceedings, the lawyers will be able to file a wide range of motions, including requests for more time to review evidence.

Browne said he thinks the U.S. government will have difficulty proving its case against Bales because "there is no crime scene" and a lack of important physical evidence like fingerprints. And he has said he wants to visit Afghanistan.

Prosecutors, meanwhile, will rely on evidence collected from the villages, including any statements by witnesses and Afghan civilians who were injured that night, as well as any observations by other soldiers about Bales' conduct and his movement on or off the base.

Two military defense attorneys also have been assigned to his case.

Ophardt said Bales' family is still living on base but has the option of moving off at any time.

Even in places as seemly well-studied as the national parks of North America, new species are still being discovered. Using ultraviolet light that cause scorpions to fluoresce a ghostly glow, researchers from the University of Nevada, Las Vegas (UNLV) have discovered an intriguing new scorpion in Death Valley National Park. They named the species Wernerius inyoensis, after the Inyo Mountains where it was found. The study was published in the open access journal ZooKeys.

This new species is small, only 16 mm in length. "We almost overlooked this one during the survey" said Matthew Graham, a PhD Candidate with the School of Life Sciences at UNLV. Matt discovered the scorpion along with his father who was volunteering that night. "Only a single male individual was found, but the physical uniqueness was enough to identify it as a new species", said Michael Webber, another PhD Candidate from UNLV who described the specimen. This new scorpion appears to be closely related to two other species found over 400 kilometers away at Joshua Tree National Park and along the lower Colorado River. This group of scorpions is most easily identified by the presence of a conspicuous spine at the base of the stinger, the function of which, if any, is unknown.

The previously known species are also rarely observed in the wild, and this elusive nature has led to speculation that these scorpions occur at very low densities or have only sporadic surface activity. However, the rocky terrain in which the previous species were found and the discovery of the new species at the base of a talus slope, hint at the possibility that these scorpions are subterrestrial, spending their lives deep in rock crevices or in the interstitial spaces among piles of loose rock.

Scorpions are quite common within arid regions where they can comprise a large component of biological diversity. The new species was discovered during field surveys funded by the National Park Service as part of efforts to develop better inventories for all organisms occurring within the parks.

"In North America, inventories for mammals, birds, reptiles and amphibians are pretty well developed, and we have a good handle on higher-order plants, but for many groups of smaller organisms taxonomic inventories will no doubt lead to numerous new discoveries" said Dr. Jef Jaeger, a Research Assistant Professor at UNLV who initiated and oversaw the scorpion surveys.

In the face of regional environmental changes brought about by human actions and the potential for larger changes that global warming may bring, many scientists and resource managers place new importance on efforts to document and catalog species diversity.

Are you on Pinterest? ?This virtual pinboard is a lot of fun, and it?s a been great tool for tracking food trends. ?The Food & Drink category on Pinterest is one of the largest and food pins are re-pinned more than pins in any other category. Food lovers are an enthusiastic bunch. ?We like to share.

  • Kale has been called the new bacon. ?I don?t know about that, but I?m sure seeing lots of pins featuring this dark leafy green ? from crispy kale chips and creamy kale smoothies to raw, massaged, marinated and braised kale in salads, soups, pastas, entrees and more.
  • Quinoa is the hot grain on pinboards. ?I?m seeing quinoa in savory pilafs, sweet breakfast porridges, salads, muffins, vegetable patties, pancakes and even desserts.
  • Sweets are a favorite of pinners, and some of the most popular themes are salted caramel, nutella and red velvet. ? If anything, red velvet seems to be the new bacon. ?Beyond red velvet cakes, cake pops and cupcakes, I?ve seen red velvet pancakes, bread pudding, cheesecake, fudge, donuts, ice cream, crepes, popcorn and marshmallows. Where will it end?
  • Popsicles may be the new cupcakes. All sorts of frozen treats on a stick are showing up on Pinterest.
  • Food in jars seems to be big, and I?ve seen pins of layered salads, vegetables and desserts in jars. ?For parties, drinks in jars are a popular trend.
  • Buffalo chicken is going crazy. ?Pins include buffalo chicken dips, appetizer bites, grilled cheese, pasta, mac? and cheese, pizza and chowder. ?Who knew there was a buffalo chicken movement brewing?
  • Other pins that I?m seeing quite often include nut butters, pesto, kimchi, mini appetizers and desserts, and meatless entrees.

LONDON - Bolton will return to the field in the English Premier League on Saturday, a week after midfielder Fabrice Muamba suffered cardiac arrest during a match and needed 15 shocks to recover.

With Muamba improving in a hospital and talking to doctors, Bolton decided to go ahead with the match Saturday against Blackburn.

Bolton team doctor Jonathan Tobin revealed Wednesday that doctors tried unsuccessfully to revive Muamba for 48 minutes before he arrived at the London Chest Hospital. It took another 30 minutes before the Muamba's heart started beating on its own after 15 shocks.

"They were working on him without his heart having a muscular beat," Tobin said. "In effect, he was dead in that time ... throughout the whole resuscitation period you are worrying.

"You know the longer the resuscitation goes on the less chance there is of survival, but this is slightly different. This is a very fit 23-year-old."

On Monday, Muamba started breathing independently again and communicating in intensive care.

"I'm glad to say that the early signs of recovery have continued," Tobin said. "I went to see Fabrice last night. I went in and he said 'Hi doc.' I asked him how he was and he said 'Fine."'

On Wednesday, New York Red Bulls striker Thierry Henry flew to London to visit his former Arsenal teammate.

Although Henry was not seen by reporters on Wednesday, Professional Footballers' Association official Bobby Barnes said the French striker had visited the hospital and Muamba.

Henry was at Arsenal when Muamba went from the academy in 2002 to the first team in 2005. The midfielder left two years later for Birmingham before joining Bolton.

Bolton manager Owen Coyle spent much of his time since Muamba collapsed at Tottenham last Saturday at the player's bedside. Coyle returned to practice on Wednesday after the game at Aston Villa on Tuesday was postponed.

"We spoke together with the players as a group this morning and I talked with Fabrice's family last night," Coyle said. "Fabrice's father Marcel and his fiancee Shauna were keen that we fulfill our fixtures. Once the players knew this, there was no doubt in our minds that we would play the matches."

Bolton also will return to Tottenham on Tuesday and replay the abandoned FA Cup quarterfinal.

It likely will be an emotional return to White Hart Lane, where Muamba suddenly fell to the ground on Saturday just before halftime. Attempts were made to revive him in front of more than 30,000 fans and a global television audience.

Dr. Andrew Deaner, a Tottenham-supporting cardiologist, leapt from his seat in the crowd and rushed on to the field to help Muamba.

"If you're going to use the term miraculous, I guess it could be used here," he said.

Opioids? molecular magic unmasked

First 3-D views show how drugs bind to cells

Proteins turned on by opium and similar substances in the body have now been caught in action. Two new snapshots show how cellular proteins lasso molecules in the opium family, revealing the 3-D structure of such pairings for the first time.?

The work represents a major step toward designing more specific analgesics and other drugs that lack opioids? nasty side effects, two teams of researchers report online March 21 in Nature.

?Both are landmark studies,? says Gavril Pasternak, a neuroscientist who designs opioids at the Sloan-Kettering Institute in New York City, and who wasn?t involved in either study. ?These structures will quickly be utilized with goal of developing nonaddicting painkillers and new ways to combat drug abuse.?

Proteins that respond to opium and opiumlike molecules protrude from the surfaces of cells throughout the brain, spinal cord and gut. The body?s own hormones and brain chemicals such as endorphins can bind to these proteins to turn the molecular switches on and off to control pain, regulate breathing and change mood.

Many of today?s most powerful painkillers work by switching on one of these proteins, called the mu opioid receptor. But the relief this provides comes at a price. Derivatives of opium, such as morphine and codeine, are addictive and can cause breathing problems and constipation.

To better understand how these drugs work, an international team of researchers for the first time crystallized a small morphinelike molecule attached to a mu receptor ? a technically difficult task that requires isolating the pair of molecules without unsticking them from each other. X-rays revealed how one molecule lined up with the other.?

This structure ?may prove to be important in understanding why some opioids are more addictive than others,? says S?bastien Granier, a molecular biologist at Stanford University and a coauthor on the mu receptor paper. Although the small molecule his team tested has the opposite effect of morphine ? it deactivates the mu receptor ? knowing how the molecule docks should help improve computer simulations of various drugs that can turn the protein on or off.

In the second study, Seva Katritch of the Scripps Research Institute in La Jolla, Calif., and colleagues looked in detail at how the experimental drug JDTic sticks to a kappa opioid receptor ? a switch that?s turned on by the plant-derived hallucinogen Salvinorin A. ?Kappa opioid receptors are especially interesting because of their emerging distinct role in regulation of stress,? says Katritch. JDTic, which flips the switch off, is being tested as a treatment for drug abuse.

Each team?s test molecule latched on to its receptor at multiple points in the revealed structures. Small differences in the attachment sites may help to explain why some molecules are more promiscuous than others. Designing new drugs that bind only with specific receptors could help to cut down on side effects.

?Everyone in the field has been waiting to see these crystal structures,? says Jane Aldrich, a medicinal chemist at the University of Kansas in Lawrence. ?Now we can look at how particular parts of the molecules interact.?

The receptor pockets that grabbed ahold of the molecules also proved to be surprisingly large and open. This extra space should give drug designers a bit of wiggle room ? and the flexibility to tack on extra atoms to drugs like morphine in the hopes of making them less problematic.

Get out the salt shaker, because there's a new Windows Phone rumor making its way across the web today. Citing sources "familiar with Sprint's LTE game plan," SprintFeed is reporting that the carrier is looking to launch a new LTE-enabled handset later this year -- one that'll allegedly run Windows Phone 8, commonly known as Apollo. These sources also claimed that the device still has to run through a series of tests, implying that it would likely hit the market this Fall, at the earliest. Beyond that, details run pretty thin, but we'll be sure to keep an eye on it going forward.
